摘要:Adenosine triphosphate (ATP) contained in an aqueous solution which alsocontains other nucleotides (e.g. AMP or ADP) as impurity is purified by (1) adsorbing theATP on to a strongly basic anion exchange resin followed by elution with an aqueous acidsolution, (2) adsorbing the ATP on to a weakly basic anion exchange resin followed byelution with an aqueous dilute alkaline solution, and (3) contacting the ATP in aqueoussolution with a non-ionic exchange decolorizing resin whereby coloring matter isadsorbed thereon while the ATP remains in solution. In carrying out the process theaqueous solution of ATP which may be in salt form (e.g. as the disodium salt) is passedthrough a strongly basic anion exchange resin, e.g. a resin of the polystyrene quaternaryammonium type, and which is conveniently in salt form, to adsorb the ATP and othernucleotides and the adsorbed ATP is preferably eluted stepwise with aqueous mineralacid eluant having different and increasing concentration of the acid (e.g. HCl) and whichmay also contain a water-soluble salt (e.g. NaCl) and the resulting fractions are then

treated with the weakly basic anion exchange resin which is preferably porous and in saltform and is suitably of the phenol-aliphatic amine type. The ATP is then eluted with, e.g.dilute NaOH solution and the resulting solution, conveniently adjusted to a pH of notmore than 5.0, e.g. 1.0 to 5.0, is passed through the decolorizing resin. Two or more resincolumns connected in series may be used in the second step to improve the adsorptionof ATP. Suitable decolorizing resins are those having a porous structure and havingphenolic hydroxy and anilino groups as active groups, e.g. those prepared by condensingphenol and metaphenylene diamine with formaldehyde. The ATP is recovered from theeffluent from the decolorizing resin and the purified solution may be treated with alower aliphatic alcohol, e.g. methanol or ethanol to crystallize the ATP, e.g. its disodiumsalt. The purified product has low toxicity and has therapeutic value.
